Epitranscriptomic RNA modifications have recently emerged to be an important layer of regulation of viral host interactions, with the gene expression of multiple viruses regulated by RNA modifications, and some modifications shielding viral RNAs from immune detection. The current COVID-19 pandemic not only highlights the importance of basic virology research, but the BioNTech and Moderna RNA vaccines also showcases the practical use of RNA modifications to prevent immune destruction of RNA vaccines.

The Tsai lab is looking for highly motivated problem solvers to join our quest in exploring epitranscriptomic RNA modifications in Influenza A virus & Human Immunodeficiency virus (HIV-1). HIV-1 projects involve international collaborations and are funded by the   Center for Structural Biology of HIV RNA (CRNA), a NIH/NIAID funded program. Potential projects include, but not limited to:

- 5-methylcytidine (m5C) regulation of Influenza A virus replication and innate immunity

- N6-methyladenosine (m6A) regulation of HIV-1 RNA stability and splicing

- RNA modification mapping using Oxford Nanopore direct RNA-seq

徵求有興趣探索病毒RNA修飾的研究夥伴一同來了解RNA修飾對病毒基因表現的調控與其延伸應用。目前已知RNA修飾可調控多種病毒的基因表現,並已有少數報告顯示RNA修飾可調控細胞分辨本體與外來病毒RNA。COVID-19疫情不但凸顯了病毒研究的重要性,BNT和Moderna的SARS-CoV-2 RNA 疫苗以人工pseudouridine修飾防止細胞攻擊疫苗中的RNA的應用例子更是RNA修飾的重要應用例子。本實驗室將針對流感病毒A (Influenza A virus, IAV) 與人體免疫缺乏病毒 (HIV-1) 兩種病毒來研究RNA修飾對病毒複製與免疫辨識的影響。詳情請參考:

潛在計畫包含:

- m5C 甲基化對流感病毒的複製與免疫之調控

- m6A 甲基化對HIV RNA穩定性與splicing之調控

- 以Nanopore RNA定序技術測定RNA修飾之位置

以上流感計畫由中研院前瞻計畫支應,而HIV計畫由美國HIV RNA結構研究中心(CRNA)支應。

實驗室常用技術包含基因轉殖、細胞與病毒培養技術、CRISPR/Cas9基因編輯、RNA蛋白質交互作用檢驗(如PAR-CLIP)與次世代定序(NGS)資料的電腦分析。參與HIV計畫者需有足夠英文溝通能力與美國合作者溝通並年年赴美國報告研究成果。若需學習CRNA合作團隊之特有技術,有機會申請經費至美國短期學術交流。

應徵資格:

Preferred candidates should be active problem solvers, either though asking collages, reading papers, or designing experiments to obtain answers. Experience with cell culture & molecular cloning needed; virology or BSL2 lab experience preferred. Must be willing to adhere to lab safety regulations.

We are currently looking for one PostDoc or Graduate level research assistant (RA with a Master's degree). RAs who wish to transition to a graduate program abroad in 2-3 years are welcome.
