針對最新的生物技術所產出的資料,本實驗室致力於發展合適的分析方法以用回答重要的生物醫學問題。近年來,蓬勃發展的高通量和酸定序的技術對於基因等遺傳物質的了解提供了全面且精準的解決方案。許多定序技術(如WGS, RNA-seq, ChIP-seq, ATAC-seq等等)目前已經被大量使用。由於比較時間序列的RNA-seq資料分析可以幫助生物學家們解開細胞裡正在發生的程序與基因的調控路徑,我們克服了分析這類資料中必須同時考慮基因表現、時間順序、與不同實驗之間的因素,發展建立了一套時序基因共表現網路(TO-GCN)方法。

這個方法已經應用在許多研究上:如人類間質幹細胞的發育、斑馬魚心臟受損再生、小鼠胚胎心臟發育、SARS-CoV-2感染人類細胞過程等,我們希望可以透過更多的合作研究,幫助生醫學者在重要或是有趣的問題上有機會可以找出前人一直以來無法得到的答案。

有鑒於單細胞核酸定序技術越來越普及,建構在原本TO-GCN的概念基礎上,我們也正發展適合分析單細胞定序資料的新方法,將分析的解析度從原本的組織層級一舉推升到細胞型態層級,分析結果更能反應真實的情況。
