[蛋白質表現差異相對定量]

Relative Protein Quantitation (TMT method)

TMT定量法是在胜肽上進行等質量同位素化學標定(isobaric tag chemical labeling )。各別樣品經過標定後混合入同一管,再利用LC-ESI/MS/MS分析,經與資料庫比對後即可同時得到蛋白質身份及其於不同樣品間的相對濃度。TMT定量法已應用於許多蛋白體研究,如細胞株、動物模式、及人體組織液,此技術可在一次實驗中進行二到八組的定量實驗,用於觀測不同樣品間蛋白質相對表現量或蛋白質轉譯後修飾程度變化。此方法適合各種複雜度之蛋白體研究,所需樣品約5ug~100ug以上,因分析程序依樣品複雜度而不同,因此欲使用此項技術請先與核心設施Dr.李福安聯絡,詳談技術細節、樣品處理步驟及預約儀器時間。目前因需預先向生化所預約高解析質譜儀,欲使用此技術者至少需於三個月前提出申請

[Relative Protein Quantitation (TMT method)]

The TMT quantitative method is to use isobaric tag chemical labeling in peptides. After labeling, each sample combined into a same tube and mixed, then analyzed by LC-ESI/MS/MS to identify the proteins and get the relative concentrations in different samples with matching to database. The TMT quantitative method has been applied to many proteomic studies, for example, cell lines, animal model, and human tissue fluid. This technique can quantitate two to eight samples at one time to observe the protein relative expression between different samples or the variation of post-translated modification. The samples should prepare more than 5ug~100ug. Please contact with Dr. Lee, Fu-An of core facility in advance for the technical details, the processing of sample treatment and making reservation for the equipment if you would like to use this technique, because the analysis program is different with the complexity of samples.
